Antibodies α-UbcM2 was raised in rabbits against a recombinant His 6 -S-tagged polypeptide corresponding to the unique N-terminal 58 residues of the enzyme (MSSDRQRSDDESPSTSSGSSDADQRDPAAPEPEEQ EERKPSATQQKKNTKLSSKTTAK), as previously described [12]. Commercially-available, validated antibodies used throughout these immunofluorescence studies include: mouse polyclonal α-GFAP (1:500; Neuromics MO22136), mouse monoclonal α-myelin CNPase (1:2500; Covance SMI-91R), mouse monoclonal α-Iba1 (1:200; EMD Millipore MABN92), mouse monoclonal α-NeuN (1:2000; EMD Millipore MAB377), mouse monoclonal α-ubiquitin (1:500; Santa Cruz sc-8017), and mouse monoclonal α-cre recombinase (1:2000; Millipore MAB3120). Secondary antibodies used were Alexa 546nm Fluor-conjugated goat α-rabbit IgG and Alexa 488nm Fluor-conjugated goat α-mouse IgG (1:1000, Molecular Probes).
